Second-generation CPIs synergize with TGFβ blockade to delay tumor growth through CD8+T cells. (A) Female BALB/c mice were injected subcutaneously (s.c.) with 2.5×10 5 syngeneic CT26 tumor cells and treated with <t>isotype-matched</t> <t>control</t> <t>IgG,</t> <t>anti-KLRG1,</t> anti-Tim3, anti-TIGIT or anti-LAG-3 antibodies (all 200 µg/dose) or the combination of those with (B, C) anti-TGFβ (100 µg/dose) starting 8 days post-tumor inoculation. Tumor growth was tracked and grouped analysis (n=10 per group) are shown. (D) Survival analysis of mice injected with CT26 tumor cells on day 0 and treated with the indicated CPIs. (E, F) Mice injected with CT26 were administered antibodies against CD4, CD8, or both starting −1 day before the start of the indicated CPI regimen and T cell depletions were continued twice a week until termination of the study. The data are represented as the mean±SEM and represent two independent experiments. P values are shown for entirety of study using two-way ANOVA test with post hoc Tukey’s multiple comparisons; **p<0.01; ****p<0.0001. Survival differences of tumor-bearing mice were assessed using Kaplan-Meier curves and analyzed by log-rank testing. ANOVA, analysis of variance; CPI, checkpoint inhibitor; PBS, phosphate-buffered saline.

Second-generation CPIs synergize with TGFβ blockade to delay tumor growth through CD8+T cells. (A) Female BALB/c mice were injected subcutaneously (s.c.) with 2.5×10 5 syngeneic CT26 tumor cells and treated with isotype-matched control IgG, anti-KLRG1, anti-Tim3, anti-TIGIT or anti-LAG-3 antibodies (all 200 µg/dose) or the combination of those with (B, C) anti-TGFβ (100 µg/dose) starting 8 days post-tumor inoculation.

Article Snippet: In vivo depletion of CD4+T cells, CD8+T cells or the combination of the two was achieved by intraperitoneal injection of anti-CD4 (clone GK1.5, RRID:AB_1107636), anti-CD8 (clone 53–6.7, RRID:AB_1107671) or isotype control IgG (RRID:AB_1107769) (all from BioXCell) with a 200 μg dose of antibody −2 days before CPI administration and a 100 μg dose every 2–3 days after that, continuing until termination of the study.
